\u0161kolskih instrumenata za fiziku i kemiju. Godine 1896. Shimadzu uspje\u0161no snima radiograf, a 1909. godine proizvodi i prvi japanski rendgen. Tijekom 1915. godine zapo\u010dinje proizvodnju opti\u010dkih mjernih instrumenta, a 1934. godine Shimadzu je razvio i prvi spektrograf u Japanu. Jednako tako, 1952. godine razvio je prvi fotoelektri\u010dni fotospektrometar, a \u010detiri godine kasnije i prvi japanski plinski kromatograf. Shimadzu je danas svjetska korporacija za proizvodnju kemijskih analiti\u010dkih i mjernih instrumenta za medicinu, industriju, zrakoplovstvo i sli\u010dno.<\/p>\n<p>&nbsp;<\/p>\n<p style=\"text-align: center;\"><a href=\"http:\/\/crowave.com\/blog\/wp-content\/uploads\/2021\/05\/hplc_dad_spdm10avp_01.gif\" rel=\"lightbox[10903]\"><img loading=\"lazy\" decoding=\"async\" class=\"aligncenter size-full wp-image-10935\" src=\"http:\/\/crowave.com\/blog\/wp-content\/uploads\/2021\/05\/hplc_dad_spdm10avp_01.gif\" alt=\"\" width=\"947\" height=\"389\" \/><\/a><span style=\"color: #00ffff;\"><em>Pojednostavljeni prikaz kompletnog HPLC sustava. Komponente spektrometra ozna\u010dene su \u017eutom bojom.<\/em> <\/span><\/p>\n<p>&nbsp;<\/p>\n<p>DAD detektor SPD-M10Avp je dio sustava HPLC koji slu\u017ei za detekciju ili otkrivanje komponenti u smjesama koje su prethodno pripremljene za takvu detekciju. Kako sama rije\u010d govori, uzorak za teku\u0107insku kromatografiju mora biti u teku\u0107em stanju (kod plinske kromatografije uzorak mora biti u plinovitom stanju). Za pretvaranje krutih uzoraka u teku\u0107u smjesu koriste se posebni sustavi HPLC-a koji uklju\u010duju otapala, visokotla\u010dne pumpe, ubrizgava\u010de i tzv. kolone gdje se odvajaju komponente uzorka primjenom razli\u010ditih fizikalnih i kemijskih parametara. Tako pripremljen uzorak iz kolone ulazi u na\u0161 detektor gdje se fotospektrometrijom mjeri prisutnost pojedine komponente.<\/p>\n<p>Princip rada DAD detektora je vrlo jednostavan. Kroz uzorak se propu\u0161ta UV ili vidljivo svjetlo, uzorak pri tome upija odre\u0111ene valne duljine svjetla karakteristi\u010dne za pojedine spojeve, te se takva svjetlost zatim raspr\u0161uje u foto-spektar na difrakcijskoj re\u0161etki kako bi se mogle izmjeriti apsorbirane spektralne frekvencije. Raspr\u0161eni foto-spektar pada na diodni senzor koji se sastoji od nekoliko stotina foto-dioda gusto poslaganih u red. Sa svake foto-diode se uzima detektirani napon koji se pretvara u digitalnu vrijednost za obradu i prikaz na ra\u010dunalu. U objavi <a href=\"http:\/\/www.crowave.com\/blog\/2020\/06\/24\/spektrometar-optical-spectrum-analyser-model-6000-6101\/\" target=\"_blank\" rel=\"noopener noreferrer\">Spektrometar Optical Spectrum Analyser Model 6000 (6101)<\/a> opisali smo sli\u010dan sistem. Temeljna razlika je \u0161to se kod tog spektrometra spektar dobivao samo preko jedne foto-diode na koju se rotirao (skenirao) odbijeni spektar. Kod DAD detektora pak nema potrebe za rotacijom jer spektar pada na niz usko poredanih dioda koje istovremeno snimaju \u010ditav spektar. Na jednoj su elementi napajanja, a na drugoj su detektorski pretvara\u010di sa ra\u010dunalom baziranom na dva 16-bitna mikroprocesora ADSP-2115 (Analog Devices) i N80C186XL12 (Intel). Ovi mikroprocesori datiraju iz 1990-tih godina pa smo na temelju toga i odredili i razdoblje proizvodnje na\u0161eg ure\u0111aja. Na izlazu su dva podatkovna SCSI su\u010delja. SCSI (<em>Small Computer System Interface<\/em>) ra\u010dunalna su\u010delja slu\u017ee za spajanje vanjskih ure\u0111aja na ra\u010dunalo i nekad su bila prisutna i na ku\u0107nim ra\u010dunalima (osobno sam imao opti\u010dki skener preko SCSI su\u010delja) no potpuno su ih stisnuli USB, FireWire i sli\u010dni moderniji priklju\u010dci. Ipak SCSI je i do danas zadr\u017ean kod nekih industrijskih ra\u010dunalnih komponenti. Deuterijska lampa radi na principu elektri\u010dnog luka. Za razliku od vodika kojem se jezgra sastoji od jednog protona, deuterij ima jezgru koja se sastoji od jednog protona i jednog neutrona. Lu\u010dne svjetiljke sa obi\u010dnim vodikom tako\u0111er imaju nagla\u0161eno zra\u010denje u UV dijelu spektra, a relativno malo u vidljivom i IC podru\u010dju. Me\u0111utim, svjetiljke sa deuterijem imaju du\u017ei \u017eivotni vijek i nekoliko puta ve\u0107i intenzitet emisije u UV dijelu spektra. Balon od kvarcnog ili UV stakla mo\u017ee s unutra\u0161nje strane biti dodatno tretiran premazom koji sprje\u010dava difuziju deutarija iz balona.<\/span><\/em><\/p>\n<p>&nbsp;<\/p>\n<p>Shema prikazuje na\u010din paljenja deuterijske lampe. Katoda u obliku grija\u010de niti zagrijava se niskim naponom (cca 4-8V\/5A) u vremenu cca 5-20 sekundi kako bi proizvela odre\u0111eni broj slobodnih elektrona za uspostavu po\u010detnog vodljivog strujnog puta prema anodi. Kad se izme\u0111u katode i anode dovede napajanje cca 300-500V preko slobodnih elektrona se uspostavi elektri\u010dni luk. Nakon \u0161to se elektri\u010dni luk uspostavio, s obzirom na ograni\u010denu struju napajanja od cca 300 mA, dolazi do pada napona napajanja na cca 100V. U tom trenutku se i smanjuje ili posve isklju\u010duje grijanje katode jer sada sam elektri\u010dni luk proizvodi dovoljno temperature za odr\u017eavanje strujnog puta. Elektri\u010dni luk pobu\u0111uje molekularni deuterij koji se nalazi u balonu lampe na vi\u0161i energetski nivo. Deuterij tada emitira svjetlost dok se vra\u0107a natrag u po\u010detno stanje. Ovaj kontinuirani ciklus je izvor neprekidnog UV zra\u010denja. Ve\u0107ina proizvedenog UV zra\u010denja koncentrirana je na jednom mjestu na maloj povr\u0161ini luka (oko 1mm) te je tu napravljen izlazni otvor (prozor\u010di\u0107) na monta\u017enom tijelu elemenata lampe. Fokusirano svjetlo sa lampi prvo se po potrebi provodi kroz filtre. Vidimo da se na okretnom postolju nalaze dva filtra (zapravo jedan prozirni filtar i jedna nepropusna prepreka), a mogu biti postavljeni tako da svjetlost ne prolazi kroz nijedan od njih (kao na slici), tako da prolazi kroz filtar ili tako da bude blokirana. Mogu\u0107e da obje lampe, za vidljivu bijelu svjetlost i za UV spektar, rade istovremeno, a pomo\u0107u filtra se po potrebi jedan od njih filtrira. Nakon filtriranja svjetlost preko ulazne le\u0107e prolazi kroz teku\u0107inski uzorak, a zatim preko izlazne le\u0107e pada na fokusno zrcalo. Sa zrcala se svjetlost preko uskog proreza ograni\u010dava glede spektralne \u0161irine, a zatim reflektira na difrakcijsku re\u0161etku koja ju rastavlja na spektar. Spektar pada na foto-diodni detektor (DAD &#8211; Diode Array Detector ili PDA &#8211; Photodiode Array Detector) u kojem se energija zra\u010denja na \u010ditavoj \u0161irini spektra istovremeno pretvara u elektri\u010dnu veli\u010dinu, koja se dalje digitalizira i obra\u0111uje u ra\u010dunalu. Ra\u010dunalo uvodi i sve potrebne korekcije izlaznog signala s obzirom na nelinearne karakteristike foto-detektora i lampe kao izvora zra\u010denja odre\u0111enog frekvencijskog spektra. Mi zbog jednostavnosti za frekvencijski spektar vidljivog i UV zra\u010denja koristimo termin svjetlost, no jasno je da se taj spektar prote\u017ee na vidljivu svjetlost i na nevidljivo UV
